PhD abstract

Since the beginning of the industrial revolution, the concentration of carbon dioxide (CO2) in the atmosphere has increased by 50% due to human activities. Each year, the ocean absorbs 26% of CO2 emissions. However, this absorption leads to ocean acidification and changes in the carbonate system. This, by affecting the stability of calcium carbonate mineral forms, affects marine biodiversity. Oceanographic measurements are essential for observing the oceanic carbonate system, supporting decision-making regarding mitigation and adaptation strategies. Monitoring acidification relies on accurate measurements of variables such as pHT and total alkalinity (TA). To ensure data quality, international recommendations and best practice guidelines have been established, such as those from the Global Ocean Acidification Observing Network (GOA-ON), which sets data quality objectives.

The thesis work aims to support oceanographers with metrological tools to ensure the comparability of pHT and TA measurement results. Metrology, including traceability, validation of measurement procedures, and uncertainty assessment, is crucial for obtaining reliable and comparable results, enabling the evaluation of long-term trends in ocean acidification.

For both parameters, reference materials have been developed and tested in inter-laboratory comparisons, allowing the evaluation of the precision and trueness of measurement methods. The uncertainty budgets of pHT and TA measurements have also been studied using two approaches of uncertainty quantification.

Finally, improvements to the metrological traceability chains are presented.

PhD abstract

Titanium dioxide (TiO2) appears to be one of the most employed pigments in the industry due to its specific structural, optical, electronic and photocatalytic properties. It is a solid, crystalline, white and bright, odourless inorganic substance in the form of a powder with particles size ranging from the nanometre (nm) to the micrometre (µm) scale. TiO2 has been used for decades as a coloring additive in food (E171), especially in candies, chewing gums and frostings. However, since the first report regarding the presence of TiO2 having particles with size < 100 nm (NPs), the concern about the toxicity of E171 increased, which led recently (2022) to an European Regulation banning the use of this additive in food in the European Union (EU). Food-grade TiO2 is, however, still used in food industry outside the EU and in food packaging all around the world, thus the question of the migration of TiO2 particles in food is raised. Reliable analytical methods for the characterisation of TiO2 are still necessary to assess human exposure by ingestion of food contaminated with E171 and TiO2-based food packaging.

The Ph.D. project addressed primarily the development and various applications of a new approach for the characterisation (detection and quantification) of NPs in complex matrices such as foodstuffs and food simulants by inductively coupled plasma-mass spectrometry in combination with the single particle approach (spICP-MS). Among the relatively large panel of available techniques for the characterisation of (nano)materials, spICP-MS shows promising features since it allows the rapid screening and analysis of the individual particles in the sample, either in terms of concentration (down to ng/L levels) and size (from ~ 30 to 900 nm).

The first part of the thesis describes the development and validation of a novel method based on spICP-MS/MS using a high efficiency sample introduction system (APEX™) for the characterisation and quantification of TiO2 NPs in food simulants. The accuracy was demonstrated for both particle size determination and quantitative measurements, and the method’s uncertainty was also calculated. The APEX™ proved to be an excellent approach to avoid matrix effects and increase the sensitivity of the method allowing the detection of particles as small as 12 nm.

The second part of the thesis consisted in the application of the developed method for two different studies. On the one hand, the APEX™-spICP-MS/MS method was used for the assessment of TiO2 NPs migration from food packaging into food simulants upon storage, showing very low migration yield of TiO2 NPs from food packaging in standard contact conditions of storage. On the other hand, the method was applied to the characterisation of TiO2 NPs from E171 in food samples. The main objective was to assess the feasibility of the method for the analysis of real food matrices, especially regarding the detection and quantification of the nanoparticulate fraction (% of particles having a diameter < 100 nm). In addition, a pilot inter-comparison study on the characterisation of E171 additive was performed using different techniques at different laboratories in France and abroad.

The last part of the thesis consisted of the development of an in-house approach (based on python code) for spICP-MS data treatment to assess the main parameters influencing the method uncertainty related to the characterisation of TiO2 NPs by spICP-MS.

PhD abstract

Sepsis is a syndrome caused by a dysregulated immune system response to an infection (bacterial, viral, fungal), leading to organ failure. In 2017, 48.9 million cases of sepsis were reported worldwide, resulting in 11 million deaths. Early and accurate diagnosis is critical for implementing a therapeutic treatment and improving patient survival. Currently, sepsis diagnosis is based on clinical evaluation of the patient, which can be influenced by nonspecific symptoms. The individual quantification of biomarkers such as procalcitonin (PCT) or C-reactive protein (CRP) using immunological methods serves as an aid in diagnosis. Since quantifying a single biomarker may lack specificity, simultaneous monitoring of a panel of biomarkers appears to be a significant advantage for early diagnosis.

In the present work, a list of nine protein biomarkers of interest for sepsis diagnosis was established in collaboration with clinicians and in vitro diagnostic manufacturers. The objective was then to develop a targeted proteomic method using high-resolution mass spectrometry (HRMS) to enable specific, sensitive, and multiplexed quantification of these biomarkers in routine, in clinical serum samples.The proteins targeted in this study have different molecular masses and serum concentrations, with a dynamic range of approximately 105 in healthy patients' serum. Thus, the quantification of these proteins by HRMS is challenging. The first step was to select two proteotypic peptides per protein, considering various criteria such as molecular mass or amino acid composition. These peptides were then used to optimise high-performance liquid chromatography conditions (chromatographic columns, mobile phases, elution gradients), ionisation, fragmentation, and HRMS analysis, as well as sample preparation workflow (protein precipitation, enzymatic digestion, peptide extraction), allowing specific and sensitive quantification of the targeted proteins. These developments led to the identification of increasingly selective protocols, resulting in a decrease in the limits of quantification. Thus, three methods were developed: i) the first targeting five proteins and meeting clinical (covering physiological and pathological concentrations) and metrological (metrological traceability) needs, ii) the second targeting eight proteins and meeting the clinical need for a highly multiplexed method, and iii) the last targeting PCT, allowing comparison with an existing reference method.To ensure reliable quantification and traceability of measurement results to SI units, a protein-based calibration was selected for proteins with existing certified reference materials (recombinant proteins. In contrast a peptide-based calibration was selected for the other proteins. Therefore, it was necessary to rigorously characterise peptide standards (purity assessment and quantification through amino acid analysis) during this work.

The validation of the first method and the evaluation of measurement uncertainties for the quantification of the five biomarkers in human serum, followed by the analysis of clinical samples, allowed the assessment of its potential implementation for early and specific diagnosis of sepsis in routine practice. Finally, the development of a robust LC-HRMS/MS method and rigorously characterised standards represents a significant advancement in establishing metrological references.

PhD abstract

For some thirty years, endocrine disruption has become a central concern at the political, regulatory, societal and scientific levels. This is due to the fact that, in the field of the environment, endocrine-disrupting compounds (EDCs), and among them, natural steroid hormones and related compounds (analogous pharmaceuticals), can exert harmful effects at concentrations below the ng.L-1 and pose a serious threat to biodiversity. In the field of human health, these compounds are widely used for the diagnosis and treatment monitoring of various diseases and disorders. The aim of this thesis was notably to ensure, in both fields, the reliable quantification of endocrine disruptors and/or biomarkers to support effective decision-making at relevant concentration levels. First, 23 steroid hormones and related compounds were selected following an extensive bibliographical work that highlighted their common interest in both fields. Then, multiresidue quantification methods by LC-MS/MS were developed in whole continental waters and human serum. Limits of quantification in the ng.L-1 and the ng.mL-1 range were respectively obtained in the field of the environment and the field of human health. Measurement uncertainty was also assessed, enhancing the degree of confidence in measurement results. Implementing the methods allowed the characterisation of many target compounds in surface waters with some of them very few documented in France and Europe. Also, traceability of measurement results to the SI was established. Present work represents a significant step towards the development of multiresidue reference methods and future certification of reference materials, which are actually limited or missing.

PhD abstract

Procalcitonin (PCT) measurement is a usefully tool to aid the early diagnosis and antibiotic stewardship for patients with sepsis. As sepsis diagnosis and antibiotic stewardship rely on fixed clinical cutoffs (threshold concentration values), ensuring accuracy and comparability of PCT results over time and space is essential to ensure an appropriate diagnostic and therapeutic management.

This thesis consisted in studying the metrological traceability of these methods of PCT measurement. The work initially consisted in documenting the current state for results’ comparability between the different commercially available immunoassays for PCT. The evaluation of correlation studies and external quality assessment schemes results indicate that properly evaluating the current state of result’s comparability and discriminating the origin of result’s discrepancies is made difficult because studies were conducted in non-harmonized conditions and commutability of the materials was not characterized. Thus, it seems essential to conduct quality external evaluation using commutable control materials. In order to evaluate accuracy of commercially available immunoassays for PCT and to meet the regulatory requirements for the metrological traceability of results, a candidate reference method based on isotopic dilution liquid chromatography coupled to mass spectrometry was developed and validated for an absolute quantification of PCT in human serum with a limit of quantification of 0.25 ng/mL. Results traceability to the SI units could be established thanks to a thorough purity assessment of the primary standards by amino acid analysis and correction of impurities contribution.

PhD abstract

The asymmetrical flow field flow (AF4) is a technique used to measure the size of nanoparticles. It consists in  fractionating the different populations inside the sample as a function of their hydrodynamic diameter. In the 1960s, a model (thereafter called classical model) has been developed, relying the retention time of a nanoparticle with its hydrodynamic diameter. However, the model validity is based on work hypothesizes which are not always respected depending on the experimental conditions. The work of this PhD thesis has consisted firstly in the study of the mechanisms governing the nanoparticles retention inside the AF4 channel. Notably it has been shown that interactions between the nanoparticles and the accumulation wall of the AF4 biased the results predicted by the classical model. Another model (thereafter called p-w model), which takes electrostatic and Van de Walls interactions into account, has been developed. Tests realized with particle standards for size showed that the p-w model give results with a better trueness than the classical model and that it can be applied for a larger range of experimental conditions. A validation of this model has been conducted and an uncertainty budget has been developed by following the Monte Carlo method. The metrological traceability of the measurement results has also been demonstrated.
